Multi-organ pathophysiology of endometriosis: Evaluating neurotrophin linked multifaceted health risks assessments

Abstract
Objective: To study Multiorgan pathophysiology of endometriosis by evaluating Neurotrophin linked Multifaceted Health Risks Assessments.
Methodology: This research analyzed 60 participants using a case-control design, including 40 endometriosis patients (mean age: 35.4±6.2 years) and 20 healthy controls (mean age: 34.8±5.9 years). This collaborative research was conducted from March to June 2023 mainly at Shaikh Zayd Hospital Lahore. We obtained samples of endometrial biopsies together with peritoneal fluid and plasma. The analysis quantified BDNF gene expression from extracted RNA through qRT-PCR testing of GAPDH as the reference gene. Clinical investigators obtained pain scores from the VAS scale and recorded rAFS classification scores for lesion severity. The statistical analysis contained both ΔΔCt calculations and a correlation analysis. p<0.05 was considered as statistically significant.
Results: The endometrial tissue showed elevated BDNF mRNA levels in comparison to eutopic endometrium (p<0.001). Endometriosis patients showed greater plasma BDNF concentrations than controls (p=0.003). Additionally, the BDNF data positively correlated with the severity of symptoms (r=0.72, p=0.001) and the extent of disease (r=0.65, p=0.004). Neurogenic inflammation received stronger validation from BDNF expression levels that increased greatly in peritoneal fluid during advanced-stage endometriosis.
Conclusion: Medical professionals can use qRT-PCR to quantify BDNF protein levels which might identify the severity of endometriosis and monitor therapeutic efficacy. Future research should focus on longitudinal studies and interventional methods to evaluate how BDNF regulation influences clinical outcomes in endometriosis and reduces associated chronic pain.
