An anti-inflammatory therapeutic perspective: Expression profiling of endodontic sealers unveiling therapeutic potential in root canal treatments

Abstract
Objective: To determine the ability of endodontic sealers in alleviating inflammation, using human periodontal ligand stem cells (HPDLSCs) and inflammatory cytokines.
Methodology: We used three root canal sealers; Endoseal MTA (E-MTA), Ketac-Endo (Ketac-E) and Sealapex (S-apex) as per manufacturer’s instructions. HPDLSCs were sub-cultured by in-vitro experiment and treated by sealers in pre-defined serial concentrations of 20ul, 40ul and 60ul, respectively based on MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] cell viability assay. Expression profiling of anti-inflammatory biometers (TNF-α, IL-1β, and IL-6) were determined via RNA extract from 24h MTT assay due to its greater reproducibility.
Result: E-MTA led to a statistically, highly significant increase in relative gene fold of Tumor Necrosis Factor (TNF-α) (3.12), Interleukins IL-1β (2.97) and IL-6 (2.77) suggesting a shift towards more anti-inflammation potential and more compatible bioavailability. Conversely, Ketac-E and S-apex diminished the level of these biomarkers, thus endorsing lesser anti-inflammatory property.
Conclusion: E-MTA possesses better anti-inflammatory effects that caused an increased gene fold of TNF- α, IL- 1β, and IL- 6. Augmentation of these effects by incorporation of bioactive glass is likely to boost these sealers’ therapeutic potential for reducing inflammation in endodontic procedures.
