Impact of sperm deoxyribonucleic acid (DNA) fragmentation on male subfertility

Abstract
Subfertility is a reproductive disease, defined as not being able to get pregnant after one year of unprotected sexual intercourse.1 Globally, it is found in 8-10% of the couples.2 Male factor subfertility has been found in 40-50% of the cases.3 Male subfertility is evaluated through conventional semen analysis but it has its own limitations and does not predict the reproductive outcome in natural conception and assisted reproductive techniques with high sensitivity and specificity.4 Normal semen parameters have been found in around 15% of the men with subfertility although it does not fully predict the integrity of sperm DNA.5,6 Sperm DNA integrity is essential for successful fertilization and embryonic development.7 Sperm DNA damage includes mitochondrial DNA damage, single and double strands DNA breaks and epigenetic abnormalities, leading to sperm DNA fragmentation (SDF).8,9
