Abstract
Objective: In-vitro analysis of antiallergic activity of AaK, (Calotropis gigantea) and Tulsi (Ocimum sanctum) for the reduction of wheat allergenic proteins.
Methodology: 14 selected wheat varieties collected from Nuclear Institute of Agriculture Tandojam (NIA) and flower of Aak and leaves & seeds of Tulsi were collected from University of Sindh Jamshoro were used for characterization using SDS-PAGE Electrophoresis
Results: Wheat proteins (allergens) after treated with extracts of Tulsi and Calotropis, showed high reduction of allergenic protein, which cause the allergic reaction to the body (specially low molecular weight protein cause allergy e.g.7 kDa, 9 kDa, 15 kDa, 12 kDa, 17 kDa, 36 kDa 45 kDa and 77 kDa). There were no allergic bands seen after treatment with Tulsi and Calotropis.

Conclusion: Wheat proteins (allergens) after treatment with extract of Tulsi (Ocimum sanctum) and Aak (Calotropis gigantea) showed reduction in banding pattern. There were no allergic band were found after treatment.

Key words: Calotropis gigantean, Ocimum sanctum, allergenic protein, wheat allergens
